What is a 24-well plate template?

A 24-well plate template is a figure of the 24-well culture plate grid — 4 rows lettered A to D and 6 columns numbered 1 to 6, with well A1 at the top left. Coloured by group with a legend it becomes a plate map: a record of which construct, treatment or control each well received.

Key facts

  • A 24-well plate has 4 rows lettered A to D and 6 columns numbered 1 to 6.
  • It is built on the same 127.76 × 85.48 mm ANSI/SLAS footprint as a 96-well plate.
  • A Corning 24 well is 15.6 mm across with 1.9 cm² of growth area — the figure Thermo Fisher gives too.
  • Corning recommends 0.38–0.57 mL of medium per well; Thermo Fisher gives 0.5–1.0 mL.

How does the 24-well plate maker work?

1

Describe the layout

Name each region by explicit ranges — untransfected cells in column 1, empty vector in column 2, construct A in columns 3–4, rows A–D as replicates. The tool knows the 4 × 6 grid and its headers; a blank request gives an empty template.

2

Pick a style

The default is a flat publication plate — white background, one outline weight, a legend. Or choose grayscale print, pastel, dark slide, hand-drawn or 3D labware.

3

Refine and export

The figure opens in your workspace, where you can recolour groups, edit the legend on the canvas, regenerate with a changed layout and export in high resolution.

24-well plate dimensions, volumes and cell numbers

The footprint is standardized; the well figures below are the manufacturers' own, so two sources are given where they differ.

Cell numbers depend on the cell line — treat the seeding and confluency figures as a starting point, and quote well dimensions from the datasheet of the plate you used.

24-well vs 12-well vs 48-well plates

The three formats share one footprint. Each step from 12 to 24 to 48 wells roughly halves the growth area per well, trading cells per condition for conditions per plate.

Where are 24-well plate figures used?

A 24-well plate holds enough cells per well for western blots, qPCR or imaging, and its map records which well got which treatment.

Transfection and knockdown experiments

Controls, empty vector and constructs in columns, replicates down the rows — the map is the record you check lysates against.

Imaging on coverslips

Wells hold coverslips for immunofluorescence; the map ties each coverslip to its condition before it is mounted.

Methods figures and protocols

A colour-coded plate figure states the layout in one panel, for reviewers and for every operator repeating the experiment.

Common questions about 24-well plates

How many rows and columns does a 24-well plate have?

Four rows lettered A to D and six columns numbered 1 to 6 — 4 × 6 = 24 wells. Wells run from A1 at the top left to D6 at the bottom right.

What is the surface area of a 24-well plate well?

About 1.9 cm², the figure both Corning and Thermo Fisher give, with a Corning well 15.6 mm across at the bottom.

How much medium goes in a 24-well plate?

Corning recommends 0.38–0.57 mL per well (3.4 mL total well volume); Thermo Fisher gives 0.5–1.0 mL of growth medium. Use the lower end for transfection mixes and the upper end for longer incubations.

How many cells should I seed in a 24-well plate?

Thermo Fisher's reference figure is about 0.05 × 10⁶ cells per well, reaching about 0.24 × 10⁶ at confluency. Adjust for your cell line's size and doubling time.

What are the dimensions of a 24-well plate?

The same 127.76 × 85.48 mm ANSI/SLAS 1-2004 footprint as a 96-well plate. The well spacing is not in the standard — ANSI/SLAS 4-2004 covers 96, 384 and 1536 wells — and is set by each manufacturer.

When should I use a 24-well plate instead of a 96-well plate?

When each condition needs more cells than a 96 well holds — protein or RNA extraction, transfection, imaging on coverslips. Use 96 wells when you need many conditions and a plate-reader readout.

Can I make a blank 24-well plate template or plate map?

Yes — ask for a blank template and you get the empty 4 × 6 grid with rows A–D and columns 1–6; describe the groups by column or well ("construct A in columns 3–4") and you get the coloured plate map with a legend.

Can it draw other plate formats?

Yes. The 384-, 96-, 48-, 12- and 6-well plates have their own pages, and any page draws another format when you name it in the description.
